The reward prediction signal
The key insight from dopamine neuroscience: the spike occurs at the cue that predicts reward, not at the reward itself. Once a reward becomes fully predictable, the dopamine signal at delivery drops to baseline. The system encodes prediction error — the gap between expected and received.
This is why novelty is intrinsically rewarding and certainty is not.
Intermittent reinforcement

When reward is uncertain — sometimes it comes, sometimes it doesn’t — the dopamine system fires most intensely. This is the neurological basis for gambling, variable-ratio reinforcement schedules, and the addictive properties of many digital interfaces. Pursuit, not possession, is what dopamine tracks.
Key pathway
The mesolimbic and mesocortical pathways originate in the ventral tegmental area and project to the nucleus accumbens (limbic/reward) and prefrontal cortex respectively. These circuits underlie motivation, wanting, and the encoding of reward prediction.
Dopamine vs. reward
Dopamine is better described as the neurotransmitter of wanting rather than liking. Animals with devastated dopamine systems will eat food placed directly in their mouth with pleasure (liking is intact) but will not work to obtain it (wanting is absent).
This distinction — wanting without liking — explains much addictive behavior: the pursuit is compulsive even when the reward itself no longer satisfies.
Adolescent dopamine
Adolescents show exaggerated dopamine responses: large rewards produce a much larger nucleus accumbens response than in adults; small rewards produce a negative response. Adolescents experience bigger-than-expected rewards more intensely and smaller-than-expected rewards as actively aversive. Peer presence triples risk-taking in adolescents but has no effect on adults — the social reward circuit amplifies everything.
